담수 환경에서의 미세플라스틱 검출 및 위해성 평가에 관한 고찰

Abstract

Microplastics (MPs), a plastic debris with a particle diameter of less than 5 mm, are a pollutant of environmental concern and have received attention in recent years. The effects of MPs on the human body and the cause of their occurrence have not been clarified, but there is no doubt that the concentration in the natural environment surrounding human life is increasing. Therefore, it is necessary to make efforts to establish the basis of MPs investigation system and risk. In the future, various researches should be carried out in order to establish scientific basis such as investigation of source and contamination status of MPs at the national level, the degree of bioaccumulation and influence of MPs emitted into aquatic ecosystem, and the harmfulness of MPs themselves. In this study, we intended to look into other studies on the collected sampling methods, pretreatment methods, and analysis methods in order to help better understand MPs in freshwater. In addition, we reviewed relating to the potential environmental risks and impacts on MPs in the freshwater environment, and proposed the risk assessment models of MPs in freshwater environments. This study will be helpful to the future research on the monitoring system, human health risk, and development of environmental policies to manage the MPs pollution.

I. 서 론

플라스틱제품은 가볍고 내구성이 뛰어나며 다목적이며 저비용으로 많은 분야에서 널리 사용되어왔다[1]. 2016년도에만 플라스틱제품의 연간 전세계 생산량은 약 3억 2천 2백만 톤이었다[2]. 그러나 최근 플라스틱 조각은 분포 범위가 넓고, 잠재적인 독성으로 인해 전세계적으로 주목을 받고 있다. Cole 등[3]이 제안한 바에 따르면 플라스틱제품의 과도한 사용량 및 생산량 증가, 그리고 잘못된 관리 등의 이유로 플라스틱 조각의 최대 10%는 해양환경에 쌓이고 있다고 한다.

2017년 9월 영국 가디언은 14개국 수돗물의 83%에서 미세플라스틱 합성섬유가 검출되었다고 발표하였는데, 미국 94.4%, 레바논 93.8%, 인도 82.4%의 순으로 검출되었고 수돗물 500mL 당 미세플라스틱 합성섬유는 1.9~4.8개가 검출되는 것으로 나타났다[4]. 이에 대응하여 우리나라 환경부도 수돗물 속의 미세플라스틱 함유 실태 조사 추진계획을 수립하고 24개 정수장의 상수원수 및 수돗물에서 미세플라스틱 함유 실태를 조사하였는데, 그 결과 상수원수 1개소에서 1개/L, 수돗물 3개소에서 0.2~0.6개/L 검출되는 것으로 조사되었다[5].

플라스틱은 환경요인에 의해 작게 조각나, 수많은 미세플라스틱으로 바뀔 수 있다[6]. 미세플라스틱의 모양과 형태는 다양하지만, 일반적으로 길이나 지름이 5mm 이하인 플라스틱 조각을 통칭하며, 1차 미세플라스틱과 2차 미세플라스틱으로 구분된다. 1차 미세플라스틱은 원래 5mm 미만의 크기로 제조된 목욕제, 화장품, 세안제 등에 포함되어 있는 플라스틱을 말한다[3]. 1차 미세플라스틱은 하천 및 수 처리장에서의 배출되어, 바람 및 지표 유출수를 통해 담수 및 해양환경으로 운송될 수 있다[7]. 2차 미세플라스틱은 일반 플라스틱으로부터 물리ㆍ화학ㆍ생물학적 작용 및 풍화작용으로 인해 마모되어 생성된 파편 및 작은 조각을 말한다[8, 9]. 2차 미세플라스틱의 오염원은 플라스틱 가방, 음식 포장지, 컵 등과 같은 소비 제품, 산업용 레진, 어구(fishing tools), 그리고 기타 폐플라스틱 파편 등이다[10]. 미세플라스틱을 구성하는 재질은 polyethylene (PE), polypropylene (PP), polystyrene (PS), polyethylene terephthalate (PET), polyvinyl chloride (PVC), polyvinyl alcohol (PVA) 등 다양하다.

미세플라스틱은 하수관을 통해 강, 바다로 흘러가기 때문에 방지 조치가 매우 어렵다. 이에 화장품과 같은 제조 분야에서 확실히 제거 또는 통제 하는 방법을 강구하고 있다. 우리나라는 2017년 4월부터 치약의 원료 또는 첨가제로 미세플라스틱을 사용할 수 없도록 하였고, 같은 해 7월부터는 이들 물질이 포함된 화장품 등의 생산 및 수입이 금지되었으며, 이어서 전면판매금지까지 되었다[11].

합성 섬유로 제작된 분해되지 않는 플라스틱은 먹이사슬의 과정을 통해서 어류, 조류에 흡수되고 결국에는 인간에게 흡수되어 인체에 영향을 미칠 수 있다. 그러나 미세플라스틱이 인체에 미치는 영향이나 발생 원인 등에 대해서는 정확히 밝혀지지는 않았다. 하지만 미세플라스틱이 인간의 삶을 둘러싼 자연에서의 농도가 높아지고 있음에는 이견이 없다. 이에 미세플라스틱 검출 조사 체계 및 위해성에 대한 근거 마련을 위한 노력이 필요한 시기이다. 1인당 플라스틱 소비량이 세계 상위권인 우리나라의 소비 패턴을 고려하여 하천 및 토양, 식수 등에서의 미세플라스틱 검출 조사를 수시로 수행할 필요가 있으며, 또한 미세플라스틱의 위해성 여부를 검증하여 사용량 감소 및 적절한 처리를 통해 일상 생활 속에서 사람들이 미세플라스틱에 대한 위험을 느끼지 않도록 조치해야 할 것이다.

그러나 미세플라스틱에 대한 국내 연구는 아주 기초적인 수준이다. 앞으로 미세플라스틱의 발생원과 오염 현황 조사, 수 생태계 내로 유입된 미세플라스틱의 생물 축적 정도와 영향, 미세플라스틱 자체의 유해성 확인, 잔류성이 큰 물질과의 흡착 정도 등 과학적인 근거 마련을 위한 다양한 연구가 우선적으로 수행되어야 할 것이다. 이에 본 연구에서는 담수 환경에서의 미세플라스틱의 체계적인 관리를 위한 분석법 확립 및 위해성 평가 방법 제안을 목표로, 문헌조사를 통해 시료채취 방법, 전처리 방법, 그리고 분석 방법 등 일관된 기준으로 종합하여 살펴본 후, 적합한 방법에 대해 고찰해 보고자 하였다. 단, 문헌 조사는 학술 데이터베이스(ScienceDirect, SCOPUS, Springer Link, KISS, RISS)에서 “미세플라스틱”과 “담수”로 검색어를 제한한 후 원문이 제공되는 문헌을 대상으로 살펴보았다.

또한 담수 환경에 존재하는 미세플라스틱의 잠재적 환경 위험 및 생물에 미치는 영향에 대한 선행 자료를 통해, 담수 환경 내 미세플라스틱의 위해도 평가 방법을 제시해 보았다.


II. 본 론
1. 미세플라스틱 시료 채취 방법 및 분석방법

미세플라스틱 분석방법은 표준화된 공인 방법은 없으나, 여러 나라에서 다양한 기관과 연구자에 의해서 연구되고 있다. 미세플라스틱의 분석 과정은 크게 시료채취, 전처리 방법, 그리고 정성 및 정량 분석 방법으로 나눌 수 있다.

1-1. 시료채취 및 보관

많은 연구자들이 시료채취를 위해 330 μm 체의 메시를 사용하는데[12-19], 이는 해양환경에서의 미세플라스틱 연구를 위해 미국의 국립해양대기청에서 제안한 333 μm의 체 눈(mesh)의 크기보다 조금 작은 크기를 사용한데 서 기인한다[20]. 다량 시료의 채취 방법은 플랑크톤 망을 설치하거나 휴대용 펌프를 이용하여 일정량의 시료를 체에 거르는 방식으로 채취하며, 소량 시료의 경우 휴대용 펌프나 양동이와 같은 도구를 사용한다. 시료 보관 용기는 플라스틱 재질보다 유리 재질이 많이 사용되며 체로 여과할 경우 회수하여 패트리 접시에 보관하기도 한다. 체의 메시 크기는 25~500 μm의 범위로 분석하고자 하는 플라스틱의 크기에 따라 다양하게 사용되고 있다[13, 21].

1-2. 시료 전처리 방법

시료에 따라 체로 거른 후에도 미세플라스틱 이외의 다양한 물질들이 존재할 수 있기 때문에, 시료 분석을 위해서는 밀도 분리 및 유기물 분해 과정의 전처리 방법을 수행한다.

먼저, 밀도 분리는 혼합되어 있는 시료 중 미세플라스틱을 다른 물질과 구분하기 위해 NaCl[20], NaI[22], ZnCl2[23] 등의 시약을 물에 녹여 밀도를 높인 후, 미세플라스틱을 상층으로 띄워 하층부로 가라앉은 물질을 제거하는 방법이다. 하지만 일부 미세플라스틱의 경우 용액의 밀도와 유사하거나 무거우면 같이 손실될 수 있다. 따라서 밀도 분리는 방해 물질이 매우 많은 경우 등 필요에 따라 수행하는 것을 추천한다.

그리고 유기물 분해는 현미경 확인 혹은 미세플라스틱에 부착된 유기물을 분해하여 측정하는 데 도움을 주기 위해 수행하는데, 유기물질을 분해하는 시약으로는 과산화수소(H2O2)가 많이 사용된다[11, 22-24]. 유기물 분해가 된 시료는 전량 회수하여 다양한 공극 크기 및 재질의 여과지로 여과하여 입자성 물질을 여과하거나 여과 과정 없이 분석하기도 한다. 여과지는 0~50℃의 온도로 건조하여 현미경 확인 및 정성 분석에 사용한다.

이외에 미세플라스틱을 다른 물질과 구분하기 위해 색소로 염색하는 전처리 방법[25], 효소 분해 방법[26, 27], 그리고 초음파 처리 방법[28, 29] 등이 있다. 효소 분해 방법은, 특정 유기물(예, 탄수화물, 단백질, 지질)과 반응을 하는 효소를 이용하는 방법으로, 효소의 값이 비싼 만큼 좀 더 최적화되어야 한다는 제한 점이 있고, 초음파 처리 방법은 부서지기 쉬운 플라스틱의 경우 전처리 과정을 통해서 미세플라스틱이 파괴되거나 더 작게 부서질 수 있다는 단점이 있다.

1-3. 정성 및 정량 분석 방법

많은 연구에서 미세플라스틱의 정성 및 정량 분석을 위해 현미경 확인과 적외선분광법을 같이 사용하고 있다. 현미경 확인은 플라스틱을 회수한 여지 위의 입자를 육안으로 확인하는데 색깔, 모양, 크기 등을 현미경이나 카메라를 연결하여 소프트웨어를 통해 확인한다. 일반적으로 현미경 확인은 실체 현미경을 40배율로 하여 사용하며[20], 이 과정에서 미세플라스틱으로 확인된 물질은 적외선분광기(FTIR spectrometer)를 이용하여 재질을 분석하는데 결과는 스펙트럼의 형태로 나타난다[23, 30]. 이 스펙트럼의 형태와 위치 등을 확인하여 미세플라스틱의 재질을 파악하는데 일반적으로 다양한 종류의 미세플라스틱 스펙트럼이 저장된 라이브러리와 비교하여 플라스틱의 재질을 확인한다.

또한 미세플라스틱을 분석을 위한 다른 방법은 라만(Raman) 분광법[31], 열분해법[32], 그리고 염색 방법[25, 33]이 있으며, 각각의 분석 원리 및 장단점에 대한 내용은 표 1에 정리하였다[21, 34, 35]. 이처럼 미세플라스틱의 정성분석 방법은 다양하게 존재하지만, 실제 환경 시료에 적용이 가능하고 정밀성과 신뢰성이 확보된, 적외선분광법(FTIR)을 많은 연구들에서 사용하고 있다[30, 36-38].

 Microscopic Counting Pretreated samples are identified by microscopy. The particles are counted directly. The particles with size down to micro-meter (mm) range can be identified by the stereomicroscope. Samples with a relative high amount of large microplastics can be identified by this methods quickly, providing an overall picture of microplastics abundance in short time with low cost. The nature of the samples cannot be determined and it is necessary to couple with identification methods listed below.
Spectroscopic
 FTIR Samples are subjected to infrared radiation with defined range and the excitable vibrations depend on the composition and molecular structure of a substance. Plastic polymers have highly specific IR spectra with distinct band patterns. Larger particle size of > 500 mm can be analyzed by ATR-FTIR. Smaller particle down to 20 mm can be analyzed by microscopy coupled FTIR. FTIR based techniques are non-destructive methods. They are well established, fast and quite reliable. The newly emerging automatic FTIR imaging such as FPA makes it possible for the fast acquisition of several thousand spectra within an area through one single measurements and the analysis time is greatly shortened. Samples must be IR reactive; the samples below 20 mm might not yield not enough absorbance interpretable spectra. Nontransparent particles are difficult to be analyzed by this method. The highly specific instruments are expensive and require experienced personals for operation and data processing. The detection is affected by the environmental matrix (e.g., biofilm formation on polymer), which creates difficulty in the data interpretation. The sample must be pretreated to eliminate IR-active water.
 Raman Spectroscopy The interaction of the irradiated laser light with the molecules and atoms of the sample results in differences in the frequency of the back-scattered light when compared to the irradiating laser. The Raman shift can be detected, leading to substance specific Raman spectra. Microscopy coupled Raman Spectroscopy (RS) method is suitable for particles size >1 mm. It is the only method available for particles falling in the range of 1 to 20 mm. Microscopy coupled RS allows the analysis for small particles between 1 to 20 mm with high spatial resolution and relatively low sensitivity towards water. It is possible to analyze nontransparent and dark particles using RS method; the fast chemical mapping can be performed by RS method, enabling fast and automatic data collection/processing. There are great interferences of fluorescence from biological, organic and inorganic impurities, hampering the identification of microplastics. Sample requires purification before analysis; appropriate Raman acquisition parameters (e.g., wavelength, laser power, and photo bleaching) are important. The automatic mapping of micro-RS is still under development. The analysis by RS is time consuming.
 Scanning Electro Spectroscopy The sample images are produced by the interaction of an electron beam with the sample to measure the secondary ions. Particle of sizes down to micro scale can be analyzed. The high-resolution image of the samples can be produced by this method. Samples need to be coated at high vacuum; no detailed identification information is available.
Chromatographic
 Thermo-analytical methods such as pyrolysis GC/MS Samples are thermally treated under ambient conditions and the released gaseous compounds are rapped, and subsequently transferred to a GC column, coupled to a quadrupole -MS. The spectra of pyrolysis products are then compared to a database of common plastic types. The method is suitable for samples with sizes > 500 mm, which can be manually handled by tweezers. The sample can be analyzed together with the organic plastic additives in one run without the use of solvents and thus avoid background contamination. The method is sensitive and reliable. Some common polymers spectra data library is available. Only one particle with certain weight can be assessed per run. The pyrolysis database is only available for selected polymers only such as PE and PP.
 Liquid Chromatography Samples are dissolved by selected solvents. Different molar mass distribution is measured by size exclusion chromatography and quantification is based on HPLC analysis. Sufficient sample size of several milligrams is required for the chemical extraction to conduct this testing. The recoveries of selected polymers were high. Inability to determine the physical characteristics, such as size information, and restrictions on polymer types limit its applications to environmental samples. Only small amount of samples can be assessed per run. Only specific polymers such as PS and PET can be analyzed by this method.
Other
 Tagging Method Hydrophobic dye adsorbs onto surfaces of microplastics and renders them fluorescent when they irradiated with blue light. Microplastics with sample size down to microscale range can be visualized and counted. This method is straightforward and allows fast screening of microplastics with low cost. The fluorescent particles can be counted and may be identified. Other particles such as organic debris might be stained by the dye. This would lead to overestimation of microplastics abundance.

그리고 정량 분석은 미세플라스틱의 개수를 세는 방식이 있으며 소프트웨어를 이용하여 크기 및 개수를 측정하여 정량하는 방법 등이 있다. 결과 표시 방법으로는 일반적으로 개/부피 혹은 무게/부피로 표현한다. 그러나 시료를 펌프나 용기를 이용하여 정체된 곳에서 정확한 취수량을 알고 채취한 경우에는 부피 당 관측 개수로 표현하는 반면, 네트를 이용하여 이동하면서 시료를 채취한 경우 면적당 관측 개수로 표현하기도 한다[1].

일반적으로 분석법의 검증에는 정도 관리를 통해 확인이 필요하다. 그러나 미세플라스틱의 경우 다양한 불확실성에 대한 검증과 방법이 표준화되어 있지 않기 때문에, 미세플라스틱의 정도 관리를 위해 일정 양의 미세플라스틱을 시료에 투입한 후 회수하는 실험 등의 연구가 진행되고 있다. 그러나 이는 미세플라스틱의 정도 관리를 위한 표준화된 방법으로 제시하기 어려우므로, 추후 많은 연구가 필요하다.

2. 담수에서의 미세플라스틱 검출 현황

미세플라스틱 연구는 주로 해양 환경에서 이루어졌고[1, 39], WEB of Science 검색 결과, 담수와 관련된 연구는 약 8% 정도(총 2,222개 논문 중 180개)였다. 표 2는 기존에 연구된 담수 환경에서의 미세플라스틱 검출량에 대해서 요약한 것이며, 단위 면적(km2)당 최고 수백만 개의 입자에서 단위 부피(m3)당 최고 십만 개의 입자까지 보고되어 큰 차이를 보이고 있다[14-18, 38, 40-51]. 이는 각 연구마다 시료 채취 방법, 시료 채취 위치, 그리고 시료 채취 지역 내 인간 활동 및 자연 조건 등의 요소들 때문이다.

담수 환경에서의 미세플라스틱 오염원은 매우 다양하지만, 가장 큰 배출원은 하수처리시설이다. 표 3은 여러 하폐수처리장에 대한 미세플라스틱 연구의 결과를 보여주며, 대부분의 하폐수처리장은 미세플라스틱을 95%까지 제거하는 것으로 나타났다[11, 52-56]. 그럼에도 불구하고, 여전히 처리장 유출수는 주변 자연수로 미세플라스틱을 상당량 배출하고 있는 것으로 나타났다[55, 57].

우리나라의 경우에도 탄천하수처리시설 방류 지점의 상류지역에서 미세플라스틱이 1.6-8.3 개/m3였음에도 하류지역에서는 7.8-12.4 개/m3로 높게 나타나[11], 하수처리시설로부터의 미세플라스틱 배출에 의해 영향을 받는 것으로 나타났다. 그러나 키카푸천 및 구스천 등 일부 하천에서는 하폐수처리시설 방류 지점의 하류지역에서 상류지역보다 미세플라스틱 농도가 낮게 나타나는 경우도 있었다[58].

3. 미세플라스틱에 의한 수생태계 영향 및 위해성 평가
3-1. 미세플라스틱에 의한 수생태계 영향

(1) 물리적 영향

물리적 영향은 주로 해양환경에 살고 있는 동물에 미치는 것으로 얽힘(entanglement)과 섭취(ingestion)로 인한 죽음이다. Laist[59]의 연구에 의하면, 매년 수백만 마리의 바닷새, 바다거북, 어류, 해양 포유류들이 플라스틱 조각을 섭취하여 소화 장애로 죽어가고 있으며, 적어도 267종의 해양생물이 플라스틱에 의해 영향을 받고 있는 것으로 나타났다.

그러나 미세플라스틱은 더 많은 생물이 많은 양을 섭취할 수 있기 때문에 수 환경에 더 큰 잠재적 위협요인으로 작용할 수 있음에도 불구하고, 아직 피해를 받고 있는 종의 수는 예측하기 어려워 과소평가되고 있을 것으로 보인다.

(2) 화학적 영향

미세플라스틱은 화학물질을 흡수할 수 있으며 먹이사슬을 통하여 농도가 상위 단계로 전달될 수 있다. Betts[60]의 자료에 따르면, 미세플라스틱의 PCBs 농도는 주변 물에 포함된 PCBs에 비하여 약 100만 배 이상 높은 농도를 나타낸다고 하였다. 또한 오래된 플라스틱이 새 플라스틱보다 오염물질을 더 많이 흡착할 수 있다고 한다[61]. 환경에서 오래 잔류하고 쉽게 이동할 수 있는 POPs 물질은 생물 축적이 가능하여, 먹이사슬을 통해 상위 단계로 전달되면서 그 농도가 점점 증가한다. 하위 단계의 생물이 POPs 물질이 흡착된 미세플라스틱을 섭취하면 상위 단계로 갈수록 고농도로 축적되며, 상위 단계의 생물의 경우 밀도가 낮아 수 표면에 부유하는 미세플라스틱을 직접 섭취할 수도 있다. 이로 인해 생식질환이나 사망을 초래하고 질병 위험을 증가시키고 호르몬 이상에 영향을 주며, 인간에게 까지 영향을 미칠 수 있다[1, 62-66].

그리고 플라스틱은 제조될 때 플라스틱의 특성을 살리기 위해 비스페놀 A, 가소제(프탈레이트), 브롬화난연제(PBDEs) 등이 첨가되는데, 이들 물질이 용출되어 영향을 미칠 수 있다[67]. 이러한 물질은 수 환경에서 플라스틱으로부터 분리될 수 있으며, 생물이 이러한 물질을 섭취했을 때 내분비계 장애 물질로 작용하며 먹이사슬을 통해 생물 축적된다. 최근 연구에 의하면, 미세플라스틱의 표면에 흡착된 오염 물질은 미세플라스틱이 섭취된 후 인체 내부에서 탈착되어 방출 될 수 있음을 보였다. 즉, 인체 소화기는 pH가 다소 낮고 온도는 비교적 높으며 소화액이 있어, 흡수된 오염 물질의 탈착 속도가 해수 등 환경에서보다 훨씬 빨라 질 수 있다는 것이다[68, 69]. 그러나 섭취된 오염 물질이 먹이사슬을 통해 각 단계에서 흡수되는 농도 및 전이되는 효율성에 대한 메커니즘은 아직까지 알려지지 않아, 추후 연구가 요구된다.

(3) 생물학적 영향

담수에서의 미세플라스틱의 생물학적 독성 영향에 대해 현재까지는 정량화된 자료가 없다. 다만, 일부 실험실 규모의 연구에서 미세플라스틱의 잠재적인 생물학적 영향에 대해 보고되었다. Ma 등[70]은 담수 생물인 Daphnia magna를 이용하여, 미세플라스틱(PS)에 흡착된 페난트렌(phenathrene)의 독성영향을 평가하였다. 그 결과, 나노 크기의 미세플라스틱은 Daphnia magna에 대해 높은 독성과 물리적 손상을 보였으며, 특히나 흡착된 페난트렌에 의해 독성이 강화됨을 보였다.

또한, 담수 어패류인 Corbicula fluninea를 대상으로 한 미세플라스틱과 항균제(florfenicol) 혼합물 독성 평가 결과, 항균제나 미세플라스틱 단일 물질에 의한 독성보다는 혼합 독성이 더 높게 나타났다[71]. 그리고 척추 동물인 제브라피쉬의 유충 및 성충을 대상으로 한 몇몇 연구에서는, 물고기가 미세플라스틱을 섭취하면 체내에서 축적이 이루어지고 그 결과, 운동 손상, 장 손상, 그리고 대사 프로파일의 변화까지 일으킨다는 것을 보여 주었다[63-66].

또 다른 생물학적 영향은 미세플라스틱의 표면에 부착되는 생물막에 의한 플라스틱의 물성 변화이다. 즉, 미세플라스틱의 밀도가 증가될 수 있다[56]. 이러한 특성으로 인해, 경질의 미세플라스틱은 수층에서 저서대로 가라 앉아 저서 생물에 영향을 미칠 수 있다. 또한, 생물막은 미세플라스틱의 표면 성질을 변화시키고 표면의 소수성을 감소시킬 수 있다[72, 73]. 그러나 아직까지 부착된 생물막이 미세플라스틱과 수계 오염물질 사이의 상호 작용을 강화시키거나 약화시킬 수 있는지 여부에 대해서는 알려지지 않아, 이 부분에 대해서는 추후 연구가 필요할 것이다.

3-2. 미세플라스틱의 위해성 평가

미세플라스틱은 전지구적으로 분포하며 환경 오염 역시 증가하는 추세에 있으므로 이에 대한 위해성 평가에 대한 연구 역시 시급하게 필요하다. 그러나 미세플라스틱의 위해성을 평가할 기존의 방법은 없다. 이에 본 연구에서는 담수 환경에서의 잠재적인 미세플라스틱의 위해성을 평가하기 위해 두 가지 측면 즉, 미세플라스틱의 검출 농도를 기반하는 방법과 미세플라스틱의 구성성분에 기반으로 한 방법을 고려해 보았다.

먼저, 오염물질의 오염도 평가를 위해 Tomlinson 등[74]이 제안한 오염부하량지수(Pollution Load Index; PLI)를 이용하는 것이다. PLI는 다른 지역들 간의 오염 정도를 비교 평가하기 위한 표준 규칙으로 간주되어 사용되고 있기에[75], 미세플라스틱의 농도 분포 및 오염도 평가를 위해서도 충분히 활용 가능하며, 아래의 공식을 이용하여 계산할 수 있다.

여기서, CFi는 미세플라스틱의 바탕 농도(Bi)에 대한 측정된 미세플라스틱의 농도(Ci) 비를 의미한다. 미세플라스틱의 바탕 농도 값은 문헌 검토 후 연구 방법, 목적 및 지리적 위치를 고려하여 인위적오염원에 의해 오염되지 않은 지역에서의 미세플라스틱의 평균 농도를 표준 수준으로 선택하여 사용한다. 이에 Bi값은 미세플라스틱 분야에 대한 더 많은 연구와 함께 더 낮은 농도가 적용될 수 있다.

다음으로 오염물질의 생태독성계수를 이용하여 특정 환경에서 다양한 오염 물질의 환경 영향을 고려하는 생태계위험지수(Ecological Risk Index; ERI)를 이용하는 방법으로, 미량 금속이 저서생태계에 미치는 영향에 대한 위해도 평가를 위해 Hakanson[76]이 제안하였으며, 다음의 공식을 이용하여 계산한다.

여기서, Ti는 Hakanson[76]에 의해 도입된 독성계수로서 독성 수준과 생물학적 민감도를 나타낸다. Pn은 미세플라스틱 시료에서 특정 중합체의 농도이며, Sn은 플라스틱 중합체의 유해 점수(hazard score)를 의미한다[77]. n은 분석한 미세플라스틱의 총 개수이다. Ei는 미세플라스틱의 독성 계수에 의한 잠재적인 생태학적 위험요소이고, ERI는 이 값의 총 합을 의미한다.

표 4는 Lithner 등[77]이 제안한 플라스틱 중합체 별 유해 점수(Sn)를 나타낸 것으로, 그들은 플라스틱 중합체의 화학적 독성이 생태계에 미치는 강한 중요한 인자로 생각하고, 각 단량체에 대한 유해 분류 및 중합체 내 단량체의 함량을 고려하여, 유해 점수를 제안하였다.

미세플라스틱의 오염 현황을 나타내 주는 CFi 값은 1.5를 기준으로 미만이면 오염되지 않은 자적인 상태로, 이상이면 인위적인 오염으로 평가되며[78], PLIERI에 대한 평가 기준은 표 5에 나타내었다[78, 79].

그러나 미세플라스틱의 위해성 평가를 위해서는 미세플라스틱의 종류, 이동 경로, 유기 오염 물질의 복합 독성, 미세플라스틱에 부착된 유해 미생물의 운반 및 생태독성학적 영향 등 포괄적인 미세플라스틱 연구사업을 통한 상시 모니터링을 추진하여 데이터를 축적하는 것이 필요하다.


III. 결 론

미세플라스틱은 수환경에서 신종 오염 물질 중 하나이다. 전세계 여러 곳에서 발견되며, 대중의 관심을 크게 불러 일으키고 있다. 우리나라의 경우 2017년 수돗물에서 미세플라스틱이 검출되면서, 산업계 배출수 및 하수처리시설 방류수 등에서 미세플라스틱 분포 실태 조사를 시작하였다. 이를 위해 국내ㆍ외 강, 호수, 및 음용수를 포함한 담수에서의 미세플라스틱 분석 연구에 대해 검토하고, 수계 영향 및 위해성 평가 방법에 대해 고찰해 보았다.

그 결과, 담수 환경에서의 미세플라스틱 검출량은, 각 연구마다 시료 채취 방법, 시료 채취 위치, 그리고 시료 채취 지역 내 인간 활동 및 자연 조건 등의 요소들로 인해, 매우 다양하였다. 또한 담수 환경에서의 미세플라스틱의 정량 및 정량화를 위한 단일 방법을 사용하는 것이 어렵다는 것을 발견하였다.

현재 우리나라는 미세플라스틱에 대한 오염현황조차 정확하게 모르며, 특히 담수와 관련된 연구는 거의 수행되고 있지않다. 따라서 미세플라스틱에 대한 조사 및 연구가 진행되면 좀 더 신뢰할 수 있는 데이터 축적을 통해 미세플라스틱에 대한 우리의 이해가 높아질 것이다. 또한 미세플라스틱을 생물이 섭취하면 체내에서 내분비계 교란 물질로 작용될 수 있고, 미세플라스틱 자체 또는 잔류성 및 생물축적성물질의 생물 축적을 돕는 수단으로써의 역할을 할 수 있기 때문에, 재활용이 어렵고 잠재적 독성이 높은 플라스틱을 중심으로 유해물질로 지정하는 등 좀 더 구체적이고 효과적인 관리 정책이 마련될 수 있을 것으로 사료된다.
